Mechanism of action

Diphenoxylate is an opiate receptor agonist that stimulates muscarinic receptors in GI, decreases the peristalsis and constricts the sphincters. Diphenoxylate has a direct effect on circular smooth muscle of the bowel and prolongs gastrointestinal transit time.
